Caspase全稱為含半胱氨酸的天冬氨酸蛋白水解酶(cysteinyl aspartate specific proteinase)。caspase是一類蛋白酶家族,成員較多,在人類,已經鑒定了11種不同的caspase,根據其蛋白酶序列的同源性可分為3個亞族:Caspase-1亞族包括 Caspase-1、4、5、11;Caspase-2亞族包括Caspase-2、9; Caspase-3亞族包括Caspase-3、6、7、8、10。起細胞凋亡執行者作用的是caspase3,6,7。

【原理】FLICA試劑FAM-YVAD-FMK(Ex/Em = 488nm/530nm),是無細胞毒性的,細胞膜滲透抑制劑。進入每個細胞,并不可逆地與活化的caspase-1結合。任何未結合的FAM-YVAD-FMK-FLICA試劑擴散出細胞并被沖走。因此留在細胞內的綠色熒光直接體現了活性caspase-1酶活性的。熒光結果可通過熒光酶標儀、熒光顯微鏡、流式細胞儀進行分析。
